چکیده

It has been reported that histones inhibit DNAdependent RNA synthesis in vitro (5, 15) and these results have been taken as support of the hypothesis (34) that histones function as regulators of gene activity in vivo. One necessary correlate of this hypothesis is that different cell types contain different amounts and/or different kinds of histones or that the histone-DNA interactions differ in different tissues. However, biochemical analyses have not conclusively demonstrated tissue specificity of histones (18). This is perhaps due to

Induced Accumulation of Nonhistone Proteins in Polytene Nuclei of Drosophila Hydei

In Diptera, the polytene chromosome puffs, regarded to be morphological manifestations of active genes (1, 2, 9), contain an increased quantity of protein if compared with their inactive state . By means of experimental induction of puff formation of certain groups of loci in Drosophila hydei, it was demonstrated that a local change in protein-dye binding capacity is one of the first events in ...

The Impression of Histone Modification and DNA Methylation in Gastric Cancer Development: Molecular Mechanism Approach

The epigenetic alterations like histone modifications , DNA methylation and others remarkable categories  including nucleosome remodeling and RNA mediated targeting have been strongly investigated  recently .In this way , beside the notable importance of DNA methylation ,the histone modifications are the most important issues in the  tumorogenesis and cancer progression. Moreover...

Structure of transcriptionally active chromatin.

Rat-liver chromatin has bee fractionated into transcriptionally active and inactive regions [Gottesfeld et al. (1974) Proc. Nat. Acad. Sci. USA 71, 2193-2197] and the distribution of nuclease-resistant complexes in these fractions has been investigated. About half of the DNA of both fractions is resistant to attack by tne endonuclease DNase II. The nuclease-resistant structures of inactive chro...

A cytological approach to the ordering of events in gene activation using the Sgs-4 locus of Drosophila melanogaster

The polytene chromosomes of Drosophila strains that differ in the synthesis of the major salivary gland glue protein sgs-4 were examined by indirect immunofluorescence using antisera to several nonhistone chromosomal proteins. The Oregon-R X chromosome, which produces sgs-4 messenger RNA, showed a strong fluorescent band at locus 3C11-12 when stained with anti-RNA polymerase II, whereas the nul...

Whole-genome bisulfite DNA sequencing of a DNMT3B mutant patient

The immunodeficiency, centromere instability and facial anomalies (ICF) syndrome is associated to mutations of the DNA methyl-transferase DNMT3B, resulting in a reduction of enzyme activity. Aberrant expression of immune system genes and hypomethylation of pericentromeric regions accompanied by chromosomal instability were determined as alterations driving the disease phenotype. However, so far...
